One man has died and police have declared a critical incident on Tuesday after a crash in western Sydney that has led to the closure of a major highway.

Motorists are being diverted due to the closure of the Great Western Highway in both directions at St Marys, between Glossop and Sydney streets.

A marked police car stopped near a dark-grey Holden Insignia sedan parked on Mamre Road at St Marys at about 4am.

The driver of the Holden then allegedly accelerated away towards the Great Western Highway and the police followed him but lost sight of the car.

The Holden then hit a silver Toyota Corolla hatchback.

The male driver of the Toyota, who was unconnected to the police activity, died at the scene.

The man has not yet been formally identified but is believed to be aged in his 40s.

The Holden driver allegedly fled from the scene on foot.

He was taken to Nepean Hospital under police guard for mandatory testing.

A crime scene has been established and a critical incident team from North Shore Police Area Command is investigating the circumstances surrounding the incident.

The investigation is also subject to an independent review.

Anyone with dashcam footage, CCTV vision or information about the incident is urged to contact police or Crime Stoppers on 1800 333 000.
